Who it is for
A team with an image-based or electrophysiology readout that produces clean-looking numbers, but no one can yet show they reflect the biology. Drug developers, CROs, imaging and instrument vendors, and academic cores buy this for the same reason: the result has to be defended, and nobody internal can say for certain where it might break.
Scope
One assay or one pipeline, end to end:
- fluorophore choice (brightness, photostability, blinking, Stokes shift), panel design and spectral overlap
- optics and acquisition
- plate layout and edge effects
- segmentation and feature extraction
- feature selection and multivariate analysis, including clustering and quantitative features
- controls and normalization
- statistics and reporting
Inputs
The protocol, your analysis scripts or vendor pipeline settings (Harmony, Columbus, MetaXpress, CellProfiler), and a representative sample — a few plates or recordings.
Deliverable
The Readout Audit Report:
- a scored rubric across roughly twelve dimensions
- a prioritized fix list, separated into quick wins and structural changes
- optionally, a re-analysis of your sample in a Python or CellProfiler notebook that you keep and can rerun (an add-on, from $4,000)
- a readout call
What it covers
Neurite outgrowth and neurite health, synaptic puncta, calcium-imaging kinetics, and MEA burst and network analysis as a data-analysis review rather than instrument service. PFF uptake and aggregation imaging, micronuclei and nuclear condensation, viability panels. ASC-speck imaging, microglia and astrocyte morphology classification with DAM and reactive-astrocyte markers, C1q and C3 deposition and engulfment, phagocytosis. Phospho-flow gating strategy review and MSD or ELISA plate QC.
Timeline
One to two weeks from the day the sample arrives.
What is excluded
Instrument service and repair, running new plates, and building a production pipeline. If the audit concludes you need a pipeline rebuilt, that is the Imaging Pipeline and Analysis Build.
